Abstract
Plant root growth was greatly influenced by the chemical properties of the soil; therefore excessive heavy metals content in the soil will obstruct the growth of root. One alternative method to overcome this problem in nickel contaminated land is through application of indigenous microbial as biological agents. The aim of this research is to study the effect of Acaulospora sp as biological agents against the growth of Canavalia ensiformis root at nickel post-mining land in Sorowako, East Luwu district, South Sulawesi province, Indonesia. The research was carried out in nursery of PT. Vale Indonesia, from December 2016 until May 2017. Randomized block design (RBD) was selected to apply treatments application of native Acaulospora sp, exotic Acaulospora sp and without Acaulospora sp as the control. Isolation of native Acaulospora sp was carried out in the laboratory of Microbiology Research and Development Hall of Environment and Forestry of Makassar, South Sulawesi. The results showed that the application of native Acaulospora sp to Canavalia ensiformis in nickel post-mine land was able to increase the number of lateral roots from 20.07 up 28.83%; root length from 11.26 to 31.76%; volume of root 20.51%; dry weight root from 5.73 to 16.77% and root-shoot ratio from 3.44 to 30.18%. The result also showed that the native Acaulospora sp give effect on acceleration process of adaptation and increase the growth of Canavalia ensiformis roots as candidate of phytoaccumulator plants in the land with high nickel concentration are latest information from this research. Further identification of native strains Acaulospora sp with high tolerance to nickel will expand the horizons of research related to bioremediation process needed in the land post-mines as land post-mine nickel in Sorowako.

REDUCING NICKEL IN SOROWAKO POST MINE AREA THROUGH APPLICATION OF MYCORRHIZA Acaulospora sp ON Canavalia ensiformis L

Abstract: The heavy metal content in area post mine nickel Sorowako reached 14.200 mg.kg-1 and became limiting factor in plant growth. Phytoaccumulator plant of Canavalia ensiformis can be inoculated with arbuscula mycorrhiza to speed up reduction process. The aim of this research is knowing ability of mycorrhiza Acalauspora sp which inoculated on Canavalia ensiformis to reduce nickel, which prepared base on randomized block design with treatment of mycorrhiza Acaulospora sp, i. e: without inoculation of Acaoluspora sp (control); inoculation of indigenous Acaulospora sp; and inoculation of non-indigenous Acaulospora sp. The research conducted in Nursery PT. Vale Indonesia, Sorowako. The results showed that Canavalia ensiformis inoculated with indigenous Acalauspora sp can was increase accumulation of nickel in plant amounted by 7,91 to 11,68% with reduction rate ranging from 2,25 to 19,02 mg.kg-1.hour-1 and absorption efficiency of 3,19 to 10,10%. The nickel concentrations in Canavalia ensiformis increased of 1.40% to 2.54%, nickel concentration in root tissue was higher than other organs at age of 30 to 60 day after planting. This result indicated that indigenous Acaulospora sp could recommendation as biological agent that can help phytoaccumulator plant at nickel post mine area in Sorowako.

Abstract Acceleration land rehabilitation management of nickel post-mining in Sorowako has been main attention of PT. Vale Indonesia. This acceleration can be done by utilizing natural resources, especially microorganism’s indigenous endomychorrhiza. endomychorrhiza also called mychorrhizal arbuscula (MA) has got a lot of attention for its ability to form a mutualistic symbiosis with 80% - 96% of plant species. This study aims to determine the dominance of spores indigenous endomychorrhiza and its potential to accelerate land rehabilitation management of nickel post-mining, which is carried out in three phases; sampling rhizosphere, trapping spores, isolation and identification of spores MA types. The results showed that the dominance of genus indigenous endomychorrhiza in the area is Acalauspora sp (75, 06%).
